Thiazolidinediones, such as pioglitazone and rosiglitazone, are oral hypoglycemic agents that increase peripheral sensitivity to insulin by binding to and stimulating the activity of nuclear transcription factors called peroxisome proliferator–activated receptors. This decreases blood glucose levels. Thiazolidinediones also decrease hepatic gluconeogenesis.
