- rb protein binds and inhibits E2F transcription factor –> prevent G1/S transition
- When ready –> Rb protein phosphorylated by cyclin depdendent kinases –> E2F is uninhibited –> cell division
- Loss of function –> retinoblastoma, osteosarcoma
